When an upload has the same name as a file already in the folder, WP Media Directories shows both files side by side and asks what to do. Replace swaps in the new file under the same attachment. Copy keeps both. Skip leaves the existing file alone.

Yes. Replace keeps the same attachment and filename, so posts and pages that use it pick up the new image.
Can I replace a JPEG with a PNG?+
No. Replace only works when the file type matches, so nothing changes type by surprise. Use Copy to upload it as a new file.
